It describes a pineal polypeptide extract from cattle, which has been found to have unique properties in in vitro and in vivo experiments [<a href=\"https:\/\/pmc.ncbi.nlm.nih.gov\/articles\/PMC11943447\/#B1-ijms-26-02691\" aria-describedby=\"B1-ijms-26-02691\" aria-expanded=\"false\">1<\/a>]. The discovery of the bioactivity of this extract and its first research were mainly led by V.K Khavinson and V.N Anisimov [<a href=\"https:\/\/pmc.ncbi.nlm.nih.gov\/articles\/PMC11943447\/#B2-ijms-26-02691\" aria-describedby=\"B2-ijms-26-02691\" aria-expanded=\"false\">2<\/a>]. Since then, Epithalamin has been widely studied, including in clinical trials, showing a normalizing effect on the basic functions of the human organism and, as a result, establishing it as a geroprotective agent [<a href=\"https:\/\/pmc.ncbi.nlm.nih.gov\/articles\/PMC11943447\/#B3-ijms-26-02691\" aria-describedby=\"B3-ijms-26-02691\" aria-expanded=\"false\">3<\/a>]. Despite the publication of several reviews of Epithalamin [<a href=\"https:\/\/pmc.ncbi.nlm.nih.gov\/articles\/PMC11943447\/#B4-ijms-26-02691\" aria-describedby=\"B4-ijms-26-02691\" aria-expanded=\"false\">4<\/a>,<a href=\"https:\/\/pmc.ncbi.nlm.nih.gov\/articles\/PMC11943447\/#B5-ijms-26-02691\" aria-describedby=\"B5-ijms-26-02691\" aria-expanded=\"false\">5<\/a>,<a href=\"https:\/\/pmc.ncbi.nlm.nih.gov\/articles\/PMC11943447\/#B6-ijms-26-02691\" aria-describedby=\"B6-ijms-26-02691\" aria-expanded=\"false\">6<\/a>,<a href=\"https:\/\/pmc.ncbi.nlm.nih.gov\/articles\/PMC11943447\/#B7-ijms-26-02691\" aria-describedby=\"B7-ijms-26-02691\" aria-expanded=\"false\">7<\/a>], to the best of our knowledge, none specifically concentrate on Epitalon.<\/p>\n<p>Epitalon, also known as Epithalon or Epithalone, is a tetrapeptide that was developed on the basis of the amino acids composition of Epithalamin [<a href=\"https:\/\/pmc.ncbi.nlm.nih.gov\/articles\/PMC11943447\/#B8-ijms-26-02691\" aria-describedby=\"B8-ijms-26-02691\" aria-expanded=\"false\">8<\/a>]. For many years, the presence of Epitalon in the human body remained unconfirmed, until 2017, when it was detected for the first time in physiological pineal gland extract [<a href=\"https:\/\/pmc.ncbi.nlm.nih.gov\/articles\/PMC11943447\/#B9-ijms-26-02691\" aria-describedby=\"B9-ijms-26-02691\" aria-expanded=\"false\">9<\/a>]. This explains why Epitalon has similar properties to Epithalamin, but differs in the intensity of some actions [<a href=\"https:\/\/pmc.ncbi.nlm.nih.gov\/articles\/PMC11943447\/#B10-ijms-26-02691\" aria-describedby=\"B10-ijms-26-02691\" aria-expanded=\"false\">10<\/a>,<a href=\"https:\/\/pmc.ncbi.nlm.nih.gov\/articles\/PMC11943447\/#B11-ijms-26-02691\" aria-describedby=\"B11-ijms-26-02691\" aria-expanded=\"false\">11<\/a>]. It should be noted that some authors do not use this common name of Epitalon and refer only to its primary structure\u2014AEDG. This name highlights the fact that Epitalon is a tetrapeptide consisting of Ala-Glu-Asp-Gly (AEDG) amino acids bound through \u03b1-peptide bonds (<a href=\"https:\/\/pmc.ncbi.nlm.nih.gov\/articles\/PMC11943447\/#ijms-26-02691-f001\">Figure 1<\/a>a). Interestingly, in one article, where the antioxidant properties of Epitalon were investigated, the authors presented a different chemical structure of Epitalon, composed of the same amino acids but bonded in a different way [<a href=\"https:\/\/pmc.ncbi.nlm.nih.gov\/articles\/PMC11943447\/#B12-ijms-26-02691\" aria-describedby=\"B12-ijms-26-02691\" aria-expanded=\"false\">12<\/a>]. In this work, the peptide bond between glutamic acid and aspartic acid was through the \u03b4-carboxylic group and the peptide bond between aspartic acid and glycine was made by the \u03b3-carboxylic group (<a href=\"https:\/\/pmc.ncbi.nlm.nih.gov\/articles\/PMC11943447\/#ijms-26-02691-f001\">Figure 1<\/a>b). The authors named this variant of Epitalon Ala\u2014\u03b3 Glu\u2014\u03b3 Asp\u2014Gly, which indicates a potential naming mistake. Embora tenha sido demonstrado que o Epitalon exerce, entre outros efeitos, uma influ\u00eancia direta na s\u00edntese de melatonina, altera os n\u00edveis de mRNA de interleucina-2, modula a atividade mitog\u00eanica dos tim\u00f3citos murinos e aumenta a atividade de v\u00e1rias enzimas, incluindo AChE, BuChE e telomerase, permanece incerto se esses s\u00e3o os \u00fanicos mecanismos de a\u00e7\u00e3o deste composto. Al\u00e9m disso, apesar do volume consider\u00e1vel de pesquisa sobre as caracter\u00edsticas biol\u00f3gicas e farmacodin\u00e2micas do Epitalon, a quantidade de investiga\u00e7\u00f5es f\u00edsico-qu\u00edmicas e estruturais deste pept\u00eddeo permanece bastante limitada.
